چکیده

2015 Approximately 30% of all currently licensed, recombinantly expressed biotherapeutic products are produced in Escherichia coli, among which a significant proportion are exported to the periplasm by the general Secretory ‘Sec’ pathway. However, this pathway cannot handle many target proteins and the Tat pathway is emerging as a powerful alternative means of export. The Tat system exports fully folded proteins, moreover it preferentially exports correctly folded proteins and has been shown to export a range of biotherapeutics. This review will discuss our current understanding of the Tat pathway and its potential application for the industrial-scale production of biotherapeutics.

Constitutive expression of Escherichia coli tat genes indicates an important role for the twin-arginine translocase during aerobic and anaerobic growth.

The transcription start sites for the tatABCD and tatE loci, encoding components of the Tat (twin-arginine translocase) protein export pathway, have been identified. Expression studies indicate that the tatABCD and tatE transcription units are expressed constitutively. Translational fusion experiments suggest that TatA is synthesized at a much higher level than the other Tat proteins.

Examination of MHC-1 Expression by J774 Macrophage Cells Treated with Recombinant Protein HIV-1 Tat 49-59 / HPV16, 18, 6, 11 E7 In Vitro

Background and purpose: Human papillomavirus (HPV) is one of the infectious agents that causes genital and non-genital warts and skin cancers in humans. The E7 protein of this virus is one of the small oncoproteins that may be the main target in therapeutic vaccines. E7 protein with HIV-1 Tat peptide (49-59), plays a protective role that cause immune Th1 and CTLs response. The aim of this study...

Novel phenotypes of Escherichia coli tat mutants revealed by global gene expression and phenotypic analysis.

The Tat protein export system serves to export folded proteins harboring an N-terminal twin arginine signal peptide across the cytoplasmic membrane. In this study, we have used gene expression profiling of Escherichia coli supported by phenotypic analysis to investigate how cells respond to a defect in the Tat pathway. Previous work has demonstrated that strains mutated in genes encoding essent...

MicroReview The Tat protein export pathway

The Tat ( twin-arginine translocation) system is a bacterial protein export pathway with the remarkable ability to transport folded proteins across the cytoplasmic membrane. Preproteins are directed to the Tat pathway by signal peptides that bear a characteristic sequence motif, which includes consecutive arginine residues. Here, we review recent progress on the characterization of the Tat syst...

Exploration of twin‐arginine translocation for expression and purification of correctly folded proteins in Escherichia coli

Historically, the general secretory (Sec) pathway of Gram-negative bacteria has served as the primary route by which heterologous proteins are delivered to the periplasm in numerous expression and engineering applications. Here we have systematically examined the twin-arginine translocation (Tat) pathway as an alternative, and possibly advantageous, secretion pathway for heterologous proteins. ...
